Comparative Evaluation Of Crude Powder Of Centella Asiatica Biology Essay

Abstraction: Centella asiatica L. ( Apiaceae ) is extremely valued for intervention of Hansen’s disease, TB, wound healing, tummy achings, arthritis, varicose venas, high blood force per unit area and as a memory foil. Centella asiatica being rich in saponins is besides reported to possess antioxidant, anticancer and antiulcer belongingss.

Triterpenoid saponins, which represent a extremely diverse category of secondary metabolites, show gastroprotective and antiulcer activity and/or utile for transition of peptic ulcers associated with Helicobacter pylori.

The major aim of the current research work was to measure the peptic ulcer mending activity of pulverization of Centella asiatica whole works and its entire saponin rich-extract and to compare their efficaciousness with the modern drug, misoprostol. Ulcer in Albino wistar rats was induced with Indomethacin ( 30 mg/kg, P. o. ) . The first dosage of Centella asiatica, its saponin rich-extract and modern drug intervention ( P. O. ) were given six hours after the indomethacin disposal and the intervention continued for the subsequent five yearss. Gastric volume, stomachic pH, free and entire sourness, ulcer index and histopathological parametric quantities were studied in control and treated groups. The decrease of ulcer index every bit good as stomachic acid end product in treated animate beings was found to be statistically important with regard to command group of animate beings. In the present research work, a suited method for optimal extraction of saponin rich-extract from Centella asiatica has been developed. Besides, a simple, rapid and sensitive UV/Visible spectrophotometric method for the finding of entire saponin content has been developed and validated. Asian acid was used as criterion for spectrophotometric measuring and entire saponin content were expressed in equivalents ( mg/g ) .

Keywords: Peptic ulcer healing, UV/Visible spectrophotometer, Indomethacin, Centella asiatica, Asiatic acid, Misoprostol.

Introduction:

Centella asiatica ( syn. Hydrocotyle asiatica ) , a works of the household Umbelliferae is a tropical medicinal works with a long history of curative usage happening in parts of India, Sri Lanka, China, Indonesia, Malaysia, Australia and Southern and Central Africa. Clinical tests have shown that infusions of C. asiatica heal lesions, Burnss and cankerous abnormalcies of the tegument, remedy tummy and duodenal ulcers, and are effectual in the intervention of Hansen’s disease, lupus, dermatosclerosis and diseases of the venas. ( Verma et al. , 1999 ; Kai et al. , 2008 ) .

Centella asiatica accumulates big measures of pentacyclic triterpenoid saponins, The biological activity of saponins has been attributed to these features. Major bioactive components among the triterpenoid saponins include madecassoside and asiaticoside, and their aglycones viz. , Asian acid and madecassic acid ( Tiwari et al. , 2010 ) . There are many bing biological and phytochemical saponin testing methods available including the vanillin-sulfuric acid ( Hiai et al.,1976, Shiau et al. , 2009 ) and antimony pentachloride colorimetric checks ( Hanzas and Barr, 1969 ; Shiau et al. , 2009 ) .In the present research work, a suited method for optimal extraction of Entire Triterpenoid saponin ( TTP ) from Centellaasiaticahas been developed and TTP content determined as Asiatic acid equivalents ( mg/g ) by UV-Visible spectrophotometer.

The major aim of the current research work was to measure the peptic ulcer mending activity of whole works pulverization of Centella asiatica, its TTP rich-extract and to compare the efficaciousness with the modern drug, Misoprostol on Albino wistarrats against Indocin induced stomachic ulceration in female albino wistar rats.

MATERIAL AND METHODS

Plant Material: Centella asiatica works stuffs were collected in the month of February, 2010 from perumbavoor, Kerala. Herbarium of the works was authenticated from Agarkar Institute, Pune. The dried works stuffs were land in a mechanical bomber and sieved through BSS sieve figure 85.

Preparation of TTP Rich extracts: The Centella asiatica works pulverization ( 50 g ) was soaked in Ethanol: Distilled H2O ( 500 milliliter ) for 1 twenty-four hours. The infusions were filtered utilizing Whatmann Filter paper no. 41 and evaporated on a H2O bath. The petroleum infusion was stored in a dessicator prior to utilize.

Vanillin-Sulfuric acid assay: For quantification of TTP from works infusions, the process used by Shiau et Al. ( 2009 ) was employed. To 0.25 milliliter of works infusion, 0.25 milliliter vanillin solution ( 10 % ) and 2.5 milliliter of sulfuric acid ( 72 % w/v ) were added and exhaustively assorted in an ice H2O bath. The mixture was so warmed in a H2O bath at 600C for 10 min and so cooled in ice-cold H2O bath. Optical density at 535 nanometer was recorded against the space utilizing a Shimadzu UV/Visible ( 160 ) spectrophotometer.

Toxicity survey: Toxicity survey for Centella asiatica works pulverization and its TTP rich infusion was performed on female albino Swiss mice as per the OECD guideline ( OECD 420, 2001 ) .

Experimental protocol for initiation of stomachic ulcers: The experimental protocol was similar to the 1 adopted by banerjee D. et al. , 2008. Female albino wistar rats weighing 180-200 g each were used in this survey. The rats were given a balanced research lab diet and given tap H2O ad libitum. They were kept at 20 ± 2 & A ; deg ; C, 65-70 % humidness, and day/ dark rhythm ( 12 h/12 H ) . The animate beings were fasted for 24 H prior to their usage in the experiments. Ulcer in the rat was induced by administering Indocin ( 18 mg/kg p.o. ) suspended in 0.5 % CMC in a individual dosage. The animate beings were deprived of nutrient for following 6 H after ulcer initiation but had free entree to tap H2O. The first dosage of centella asiatica works pulverization, its TTP Rich infusion and misoprostol were given 6 H after the indomethacin disposal. In the subsequent six yearss, the trial samples were given at 9 AM on each twenty-four hours. The rats were sacrificed on the 1st, 3rd and 7th yearss, 4 hour after administrating of the last dosage.

Determination of stomachic juice volume and pH: The stomachic juice volume and pH was determined as described by Zaman R. et Al.

Macroscopic rating and stomachic ulcer index: The stomachic ulcer index was calculated as described by Dokmeci D. et Al.

TPP Rich infusion: The optimum conditions for the extraction of Entire triterpenoid saponins contained in Centella asiatica works pulverization include the undermentioned parametric quantities:

The volume fraction of Ethanol:50 %

Extraction clip:12 hour

Solvents to raw stuff ratio 20:1.

Under these conditions, the experimental output was 15.74 mg/g

Decision:

Overall, the present survey established that Centella asiatica and its TPP rich infusion possesses important mending belongings against Indocin induced tummy ulceration in rats. These consequences along with its non-toxicity suggested PK as a promising anti-ulcerogenic preparation for farther rating.
